Glycerol plays multi-functional roles in cellular physiology. Other than forming the backbone molecule for glycerophospholipid and triglyceride (TG), glycerol acts as an energy substrate for glycolysis. Spermatozoa are known to utilize glycerol for energy production, but there are no reports of this in oocytes. In this study, the value of glycerol as an energy substrate for bovine oocyte maturation (Exp. 1) and the gene expression of glycerol kinase (GK), an enzyme crucial for cellular glycerol utilization, in bovine oocytes and early embryos (Exp. 2) were examined. Special attention have been paid in members of the transforming growth factor (TGF-β) superfamily; growth differentiation factor 9 (GDF9/gdf9) and bone morphogenetic protein 15 (BMP15/bmp15), which exert regulatory functions during oocyte maturation and follicle development. Cigarette smoke is known to be a serious health risk factor and considered reproductively toxic. In the current study, we investigated whether constituents of cigarette smoke, pyrazine, 2-ethylpyridine, and 3-ethylpyridine, adversely affect reproductive functioning such as oocyte maturation and sperm capacitation. Our findings indicated that three smoke components were involved in retardation of oocyte maturation in a dose-dependent manner and the lowest-observed-adverse-effect level (LOAEL) was determined to be 10-10M. INFLUENCE OF HORMONES AND FOLLICULAR FLUID ON MATURATION OF PIG OOCYTES Ciência Rural

To evaluate the effect of follicular fluid on in vitro maturation, pig oocytes were cultured in the presence of hormones where 10% of fetal calf serum (FCS), 10% of follicular fluid from large follicles (l-pFF), 10% of follicular fluid from medium follicles (m-pFF) or no supplement were added. When oocytes where matured in medium containing the hormones the addition of different supplements did not affect (P<0.05) nuclear maturation. However, changing the supplement altered the cytoplasmic maturation, with higher rate (P<0.05) observed in the l-pFF group.
